Induction of apoptosis and cell cycle arrest in glioma cells by GL331 (a topoisomerase II inhibitor).

چکیده
BACKGROUND GL331 is a topoisomerase II inhibitor and, in this study, the effects of GL331 upon rat C6 glioma cells were investigated. MATERIALS AND METHODS The glioma cells were treated with GL331, then a cytotoxicity assay was done to evaluate the cytotoxic effects of GL331 and flow-activated cell sorter analysis was performed to analyze the cellular apoptosis and cell cycle distribution of the glioma cells. The expressions of p53, p21 and 14-3-3sigma in glioma cells treated with GL331 were investigated by Western blot analysis. RESULTS GL331 was demonstrated to exert cytotoxic effects upon and induce apoptosis of the C6 glioma cells in a concentration- and time-dependent manner (p < 0.05). GL331 also caused the glioma cells to accumulate in the G2/M-phase of the cell cycle and increased the expressions of p53, p21 and 14-3-3sigma. CONCLUSION GL331 exerts cytotoxic effects and induces apoptosis in C6 glioma cells, accompanied by cell accumulation in the G2/M-phase and activation of p53, p21 and 14-3-3sigma.

Radiosensitizing effect of deferoxamine on human glioma cells
ABSTRACT Background: Tumor cells exhibit an increased requirement for iron to support their rapid proliferation. Deferoxamine (DFO), an iron chelator, has been reported to have anti-proliferative effects on cancer cells through induction of apoptosis and cell cycle arrest. X-rays also induce apoptosis and cell cycle arrest. However, limited information is available regarding the effect of iron...
متن کاملG1 Phase Arrest and Apoptosis Induction in Human Thyroid Cancer Cell Line Thr.C1.PI33 by 3-Hydrogenkwadaphnin Isolated from Dendrostellera lessertii
Dendrostellera lessertii (Thymelaeaceae) is a toxic plant that grows in parts of Iran. The anti-proliferative properties of its crude methanol extract and one of its active components, 3-hydrogenkwadaphnin (3-HK), have been established using several cancer cell lines. Methods: In a further attempt to determine the mode of action, two groups of synchronously growing cells were treated with a sin...
متن کاملThe topoisomerase II inhibitor, genistein, induces G2/M arrest and apoptosis in human malignant glioma cell lines.
The protein tyrosine kinase inhibitor, genistein, has been reported to inhibit proliferation and to induce cell death in various non-solid and solid cancer cell lines. Herein, we examined the effects of genistein in several human malignant glioma cell lines. We found that genistein inhibited the proliferation of LN-18, LNT-229, LN-308 and T98G cells at EC50 concentrations of 25-80 microM (72 h ...
متن کاملCombination of Ethanolic Extract of Citrus aurantifolia Peels with Doxorubicin Modulate Cell Cycle and Increase Apoptosis Induction on MCF-7 Cells
New approach of breast cancer therapy is developed toward combination therapy with agent that have a specific molecular target. Our previous study showed that Citrus aurantifolia lime peels ethanolic extract (CPE) increased the sensitivity of MCF-7 cells againts doxorubicin. This study aims to observe the mechanism of combination CPE and doxorubicin in cell cycle modulation and apoptosis on MCF...
متن کاملERK1/2 Signaling Plays an Important Role in Topoisomerase II Poison-Induced G2/M Checkpoint Activation
Topo II poisons, which target topoisomerase II (topo II) to generate enzyme mediated DNA damage, have been commonly used for anti-cancer treatment. While clinical evidence demonstrate a capability of topo II poisons in inducing apoptosis in cancer cells, accumulating evidence also show that topo II poison treatment frequently results in cell cycle arrest in cancer cells, which was associated wi...
